Dissecting Proton Delocalization in an Enzyme’s Hydrogen Bond Network with Unnatural Amino Acids

Wu, Y.; Fried, S. D.; Boxer, S. G. Biochemistry 2015, 54, 7110–7119. [DOI]

Unnatural amino acids make it possible to systematically tune the extent of proton delocalization in a hydrogen bond network by altering the pKa’s of the participating residues. The characterization of this H-bond network in the resulting unnatural mutants illustrates the structural and chemical requirements for efficient proton delocalization.
