Note: You will need to design sequencing primers and order them before sending your samples out for sequencing.

  1.  Dilute the 100 uM primer stock to 5 uM (95 uL water + 5 uL stock).
  2. Add 2.5 uL primer to 5 uL template if the template is at (50-100 ng/uL).
    1. If the template concentration is much greater than 100 ng/uL, dilute to ~100 ng/uL.
    2. If the template concentration is less than 50 ng/uL, use 5 uL undiluted.
  3. Fill out the GeneWiz Sequencing form as an Excel file, save for your records.
    1. Make sure that the DNA name matches the tube and SnapGene file.
    2. Leave concentration blank.
  4. Go to GeneWiz.com>Login>Sanger Sequencing>Plasmid (or whatever you are sequencing)
  5. Service type: Premixed
  6. Service priority: Standard
  7. Upload your GeneWiz Sequencing form
  8. Order name: Date
  9. Save and review>order>add to cart
  10. Insert address information (once)
  11. Sample submission: dropbox
  12. Create a new PO: 09182018_8XXXXXXXXX_AF (Date_budget#_initials)
  13. Checkout
  14. Print order confirmation page and save PDF
