Tagging and Enriching Proteins Enables Cell-Specific Proteomics

Elliott, T. S.; Biano, A.; Townsley, F. M.; Fried, S. D.; Chin, J. W. Cell Chem. Biol. 2016, 23, 805-815. [DOI]

We demonstrate a method that applies inverse electron-demand Diels Alder chemistry to efficiently label proteins that contain an unnatural amino acid, allowing those proteins to be selectively isolated from a crude lysate. This allows us to identify low-abundance proteins in E. coli and perform cell-specific proteomics in Drosophila.

A perspective by Haoxing Wu and Neal Devaraj does a great job of explaining the work to a more general scientific audience.
