We demonstrate a method that applies inverse electron-demand Diels Alder chemistry to efficiently label proteins that contain an unnatural amino acid, allowing those proteins to be selectively isolated from a crude lysate. This allows us to identify low-abundance proteins in E. coli and perform cell-specific proteomics in Drosophila.
